Electron Microscopy Image Processing
This platform will provide personalized support for Instruct projects requiring specialized processing of cryo-electron microscopy (cryo-EM) images. Starting from either raw movies or aligned micrographs, a complete Single Particle Analysis (SPA) workflow is applied using state-of-the-art software packages (Relion, cryoSPARC, Xmipp, cisTEM, etc.) integrated into Scipion. The goal is to obtain a cryo-EM map (or a set of maps) at a resolution suitable for protein modeling. The platform will also assist developers in integrating their methods into widely accessible analysis workflows. The Instruct Image Processing Centre (I2PC) in Madrid will provide personnel for this analysis, along with the necessary computational resources.

In this service, users are encouraged to provide a particle set that has undergone prior 2D and 3D classification but still exhibits a certain degree of flexibility, either compositional or conformational. The dataset's conformational landscape is then thoroughly analyzed using state-of-the-art software packages such as Zernikes3D, cryoDRGN, opusDSD, cryoSPARC 3DVA, and more, in combination with flexible reconstruction methods to correct for motion, including Relion5 - DynaMight, cryoSPARC - 3D Flex, and Xmipp - ZART. This multi-algorithmic approach is facilitated by Scipion, a cryo-EM image processing framework developed by our team, which integrates various tools into a single platform.
